Dynamic modelling of l-threonine production by e. coli using whey and fish hydrolysates as substrates
L-Threonine is an essential amino acid widely used in food, animal feed, cosmetics and medicine, which is mainly produced by microbial fermentation. In this work, the potential of L-threonine production from a culture media previously designed that uses agricultural and fishery wastes is studied at bioreactor scale (7.5L). Batch and fed-batch experiments were carried out using whey as carbon source and Red Tilapia (Oreochromis sp.) viscera hydrolysates, as nitrogen source. The experimental data obtained were used for developing a low-structure kinetic model which comprises the mass balances for the metabolites and 34 parameters that were identified. A two-step procedure for parameter identification was implemented. First, a general set of parameters (independently of the operating mode) was found. Second, the 10-most sensitive parameters were identified, specifically for each operating mode. The model proposed in this work allowed to properly describe the kinetics of biomass production, sugar consumption and L-threonine production, and it will be used in a future for the optimization and control of the bioprocess.
